Design Notes

The part includes sacB-5, tac promoter, pgi gene, rrnB-T terminator, sacB-3. sacB-5 and sacB-3 are the flanking sequences of the sacB gene. Through the action of the bacterium's own homologous recombination enzymes, the two homologous arms facilitate the exchange, allowing the pgi gene expression frame to replace the location of the sacB gene.
